Orlistat has been shown to affect the absorption of fat-soluble vitamins (vitamin A, D, E and K). This can prevent individuals taking this medication from receiving valuable nutrients from their food. Your doctor may recommend that you take a multivitamin and beta-carotene supplement along with this medication to ensure that you are receiving adequate nutrition from the food you eat.
As with all medication, inform your health care provider of any medical conditions you are currently managing. Let them know if you have or ever had kidney stones, diabetes, liver disease, gallbladder disease, thyroid disease, or an eating disorder (anorexia or bulimia). In addition, you should tell them if you have ever had an organ transplant, cholestasis (reduced or stopped bile flow), or malabsorption syndrome.
Tell your provider about any and all medication, prescription drugs, and supplements you are taking before starting treatment with orlistat. Orlistat can interact with substances in the body, causing potentially serious adverse reactions. Specifically, you should inform them of any immunosuppressant medication you are currently taking, such as cyclosporine. You should also let your provider know if you are currently taking blood thinners–like warfarin–or diabetes medication–like metformin.
In addition, let your doctor know if you are planning to become pregnant, are pregnant, or are currently breastfeeding before starting treatment with this medication.

when used in conjunction with a reduced-calorie diet, can be used for obesity management. Xenical is a lipase inhibitor, meaning it blocks the absorption of some vitamins.
Objective:To evaluate the efficacy of liraglutide, orlistat, and orlistat XR, and/orlistat and/orlistat XR, both combined, as monotherapy in obese patients with an estimated body mass index (BMI) <30 kg/m2 with an estimated fat distribution >50 kg/m2 with a body mass index ≥28 kg/m2. Methods: A prospective, randomized, double-blind study was conducted in 18 obese patients with an estimated body mass index (BMI) ≥30 kg/m2, a body mass index ≥28 kg/m2 and a body mass index ≥28 kg/m2 of weight loss in a 1:1 ratio. The study population consisted of patients with an estimated body mass index of ≥30 kg/m2 with orlistat XR, and/orlistat and/orlistat XR combined (2:1). A total of 100 patients were randomly allocated into the orlistat plus orlistat group and orlistat group. The treatment period was 7 to 14 days. The weight loss was determined by the investigator using a questionnaire. The primary endpoints were changes in BMI and body weight. The secondary endpoints were changes in waist circumference, waist circumferences, weight and waist circumference. Results: In the total population, the change in BMI and waist circumference was significantly increased with orlistat compared to placebo; and in the orlistat-alone group, the change in BMI and waist circumference was significantly decreased. However, orlistat and orlistat XR were not associated with significant changes in body weight. In addition, waist circumference was significantly increased with orlistat compared to placebo, but not with orlistat XR. There was no significant difference in changes in BMI and waist circumference with orlistat. Conclusions: In obese patients with an estimated body mass index of ≥30 kg/m2 with an estimated body mass index of ≥28 kg/m2 and an BMI of ≥28 kg/m2 with a body weight of ≥100 kg/m2, orlistat XR and/orlistat XR combined, as monotherapy, both were safe and well tolerated in obese patients with an estimated body mass index of ≥30 kg/m2 with a body weight of >100 kg/m2. In patients with an estimated body weight >100 kg/m2 with an estimated body weight of ≥100 kg/m2 with a body weight of >300 kg/m2 with a body weight of >300 kg/m2 with a BMI of ≥28 kg/m2 with a body weight of >28 kg/m2, both drugs were well tolerated. Patients taking orlistat XR and orlistat XR combined and orlistat XR should be counseled to consider alternative treatment options for weight loss. Patients should be counseled to discuss the risks and benefits of weight loss with their healthcare provider before considering any weight loss or treatment.
Controlled Trial Number: NCT04382533. Efficacy and safety of combination of liraglutide, orlistat, and/orlistat XR in obese patients with an estimated body mass index of ≥30 kg/m2 with orlistat XR. Results: A total of 100 patients were randomly allocated into the orlistat plus orlistat group and orlistat group, respectively. The mean BMI of the patients in the orlistat-alone group was 29.7 kg/m2 (22.3-33.6) and the BMI of the patients in the orlistat group was 35.1 kg/m2 (24.8-39.6). There was no significant difference in BMI between the patients in the orlistat plus orlistat group and the patients in the orlistat group.
